Neuwirth was born on December 31, 1958, in Newark, New Jersey, United States to her mother Sydney Anne, and her father Lee Paul Neuwirth. She grew up with her older brother Peter. Neuwirth began taking ballet lessons at five, a year after viewing a production of The Nutcracker with her mother.

However, after viewing the musical Pippin in Manhattan at 13, she changed her plans from becoming a ballerina to being a Broadway musical dancer. After graduating from Princeton High School in 1976, Neuwirth went to the Juilliard School for dance and dropped out after only a year, disliking the school for not having Broadway-style dance training.

Soon after, Neuwirth took singing and jazz classes at a New York City-based YWCA, where she performed with the Princeton Ballet Company in Peter and the Wolf, The Nutcracker, and Coppélia.

Neuwirth rose to fame for her part on stage and screen. She has received a Drama Desk Award, two Emmy Awards, and two Tony Awards. In 1980, Neuwirth debuted on Broadway in the musical A Chorus Line.
