Mangione was born and brought up in Rochester, New York, United States, to Italian parents. His paternal uncle was author Jerre Mangione. With his pianist brother Gap, they showed the Mangione Brothers Sextet/Quintet, which recorded three albums for Riverside Records, before Mangione branched out into other work.

Chuck studied at the Eastman School of Music from 1958 to 1963, then entered Art Blakey’s Jazz Messengers, for which he served as the trumpet chair formerly held by Clifford Brown, Freddie Hubbard, Kenny Dorham, Bill Hardman, along Lee Morgan.
