Glenister was born in Harrow, Middlesex, England, on February 10, 1963. He is the eldest son of director John Glenister and Joan Fry Lewis, and the younger brother of Robert Glenister, a fellow actor.

Glenister went to Hatch End High School. In his early profession, he performed as a runner for the Robert Stigwood Organisation. Subsequently, Glenister worked as a movie publicist though he developed an interest in acting at the age of 23 after watching his younger brother perform in a student play about the Sex Pistols.
